    Given the reproductive difficulties that differences in plant ploidy may cause in the movement or mixture of plant populations, our group is considering incoporating analysis of plant ploidy into a grant evaluating genetic structure of plant populations. 

    Most of the protocols for determining plant ploidy via flow cytometry require fresh plant tissue that should be stored on ice until laboratory processing. Permit constraints and past tissue sampling would make fresh sampling much less feasible than analysis of the dry samples we already have in hand. I found a paper from 2006 that advocates the use of DAPI flow cytometry for determining ploidy for dry tissues (see link below or file attached). Does anyone have any experience with measuring plant ploidy in fresh or dry tissue that might be able to shed light on this?
