The necessity of tissue culture depends on the propagule that is available to you. Pollen and whole seeds may sometimes be preserved and recovered without in vitro. Vegetative structures (such as meristem, shoots, and buds) need to be cultivated and recovered using micropropagation. Some seeds may need to have the embryo removed to cryopreserve and recovered in vitro.
